Just to be sure, if the block quote contains some italic text {\it ...} and some emphasized text {\em ...}, you want the emphasized text to come out differently from the italic text. Is it okay to use a macro \emph{...} instead of a font switch {\em ...}?

In your last example you mention that you also want the "block quote regular font" to change from Times New Roman to Liberation Serif. That would imply changing the entire blockquote style

You can define two typefaces (or two font families) and add \setupdelimitedtext[blockquote][style={\switchtobodyfont[other]}].

Works here with ConTeXt MkIV 2015.10.09. However, if you are starting from scratch, I would suggest that you use the Natural Table environment rather than tabulate.

The \setuphead command does not take spacebefore and spaceafter keys. You have to set these space using before={\blank[force,1cm]}, after={\blank[3cm]}, etc.

@Manuel: The idea is that inmargin places the bullet in the ... well ... the margin. If you just want a negative left kern, you can use \startitemize[margin=-2em, width=2em], etc. See my updated answer

paragraph environments do not use before and after keys. They are primarily meant to tag paragraphs for XML/XHTML export. If you need something to alter the display, why not use framedtext (that has the advantage of not splitting across pages?
