If I understand you correctly you are searching for the primitive \inputlineno:

The fancyvrb package lets you print line numbers along with verbatim material, and it lets you customize how the line numbers are formatted.
\documentclass{article}
\usepackage{fancyvrb} % for 'Verbatim' environment

\renewcommand{\theFancyVerbLine}{{\scriptsize Line \arabic{FancyVerbLine}}}

You'll have to change the formatting of the numbers to be circled:
\documentclass{article}
\usepackage{algorithm,algorithmic}
\usepackage{circledsteps}% https://tex.stackexchange.com/q/7032/5764
\newcommand{\circlenum}[3]{\Circled{\small #1{#2}}}
\algsetup{linenosize=\circlenum}

Since you don't want line numbers, the simplest method is to disable them altogether:
\documentclass[journal,article,submit,moreauthors,pdftex]{Definitions/mdpi}
\renewcommand{\linenumbers}{}

On 14th December 2020, MDPI has updated its LaTeX template. This command
\preto{\abstractkeywords}{\nolinenumbers}
is not working. In the new template, the page number starts after the template line
